(A) The orientation of a thermotropic LC such as 5CB is fixed at a glass surface. A polariser is placed below the set-up (shown as a thick black line). Selection of one plane of polarised light by the polariser allows that light wave to propagate through the favourably aligned LC and appear at the top as a uniform area of brightness. (B) The model protein is immobilised onto the lower glass surface. However, the alignment of the LC is disrupted around the biomolecule resulting in local isotropy. Light waves are unable to propagating through this area, leading to the appearance of a dark ‘patch’ in the otherwise uniformly bright area. (C) A larger biomolecule such as a cell or cell aggregate is ‘free-floating’ in a lyotropic LC system, causing a local change in anisotropy—which appears as a dark patch.
